After Moderna’s $44B Oncology Win, Silexion’s Pancreatic Cancer Treatment is Worth Exploring (NASDAQ: SLXN)

Moderna's cancer news sent its stock up 177% in a single day (NASDAQ: MRNA), from $62.96 to $174.38. Roughly $44 billion of market value was added in one session, while Merck rose 12.6% to a record high. The reason was simple: a large Phase 3 study showed that Moderna's personalized cancer vaccine worked better with Merck's Keytruda (NYSE: MRK) than Keytruda alone at keeping high-risk melanoma from coming back or spreading.

That was a huge result for Moderna, but it also sent a simple message across biotech: helping a blockbuster cancer drug work better can be enormously valuable. Keytruda generated $31.7 billion in sales in 2025 and is one of the biggest cancer drugs in the world. Moderna did not replace it. It added something new and made the overall treatment more effective. That is where Silexion Therapeutics (NASDAQ: SLXN) starts to get interesting. Why Pancreatic Cancer Is So Hard

By shutting down the message that produces the faulty KRAS protein, Silexion hopes to slow or stop that growth across several common KRAS mutations.

Silexion's New Immune Angle

The newer part of the story is what SIL204 may do to the tumor itself. In recent lab studies, Silexion saw changes in several immune signals tied to how cancer cells are recognized, attacked and able to hide from the immune system. Put simply, the treated cancer cells appeared easier for the immune system to see and more vulnerable to being killed.

That matters because pancreatic cancer is especially difficult for immune-based drugs. If SIL204 can both attack KRAS and make the cancer easier for the immune system to recognize, it could open another path for the program beyond its current use with chemotherapy. Silexion has already said the findings support eventually studying SIL204 together with drugs like Keytruda, though that combination has not been tested in patients and the supporting data so far are preclinical.

Why the Moderna Read-Through Matters

Moderna and Silexion use different technologies, but the bigger opportunity is easy to understand. Moderna just showed how valuable it can be when a new cancer treatment makes Keytruda work better. Outside research has also shown that shutting down KRAS can help immune cells attack pancreatic tumors more effectively, which gives Silexion a reason to explore the same combination idea from a different direction.

That gives SIL204 two paths for investors to watch. The first is the one Silexion is already pursuing: attacking KRAS-driven pancreatic cancer. The second would be a future immunotherapy combination, where SIL204 could help turn a tumor that hides from the immune system into one that drugs like Keytruda can attack more effectively. That could broaden the opportunity around the same drug without changing the core KRAS story.

The Clinical Program Is Already Moving

Silexion is also moving beyond the lab. On July 29, the company activated Tel Aviv Sourasky Medical Center, known as Ichilov, as the first site in its Phase 2/3 study of SIL204 in locally advanced pancreatic cancer. Patient screening is cleared to begin, and additional centers in Israel and Germany are moving toward activation. The study starts with about 18 patients for safety before expanding into a randomized group of about 166 patients.

The current trial is testing SIL204 with standard chemotherapy, not Keytruda, so the immunotherapy angle is a future opportunity rather than part of the study now underway. But the basic setup is clear: Silexion already has a Phase 2/3 pancreatic cancer program moving into patients, while new lab data are opening a second possible use for the same drug. Moderna's $44 billion one-day move showed how quickly the market can react when a new cancer approach helps an established treatment work better. Silexion now has to show what SIL204 can do in patients.
